(A-216) Exploring and Characterizing Glioblastoma Data from a Real-world Clinical Platform

Background: Secondary real-world data (RWD) sources are pivotal in enhancing our understanding of cancer treatment and outcomes outside of clinical trial settings. There are a variety of specialty oncology RWD data sources; however, richness of data varies across these data sources.

Objectives: To describe data available in a direct-to-patient, real-world decentralized precision oncology plat¬form based on electronic medical records and determine its fitness for research purposes.

Methods: Using xCures clinical data curated from medical records of patients with any record of glioblastoma diagnosis (≥1 SNOMED code, ICD-O-3 code, or free-text term) between 2012-2024 and with availability of specific data elements, we described the distribution of demographic characteristics, comorbid conditions, procedures, medications, cognitive performance exam record availability and results, and mortality events using Aetion® Substantiate. No additional exclusion criteria were applied. Patients were followed up through the first of death, patient data end, or data end (2024-01-17).

Results: Among the 282 patients included, the median [IQR] age was 55 [45, 62]. Patients were predominantly male (63.5%) and White (80.5%), with notable missingness in race (12.4%) and ethnicity (19.9%) data. Of the 199 (70.6%) patients with insurance data available, 65.2% were commercially insured. Almost all patients had a record of a brain MRI (95.0%) and were taking medications (99.6%) over follow-up. The most common medications were temozolomide (79.8%), dexamethasone (65.4%), and levetiracetam (50.0%). Performance exams occurred more frequently as the disease progressed, increasing from 28% in the 14 days following glioblastoma diagnosis to 89.0% during the entire follow-up period. The means (sd) of Karnofsky Performance Status and ECOG Performance Status scores over follow-up were 74.21 (18.52) and 1.34 (1.19), respectively.

Conclusions: Given completeness and relevancy of the conditions and procedures data elements, xCures data appears fit for describing and better understanding target populations; however, broader utility remains to be determined.
